Study on the Changes of Surfactant Protein A from SD Rats with Pulmonary Fibrosis Induced by Bleomycin Injection

Abstract
Objective: This study was conducted to evaluate the changes of surfactant protein A in the bronchial washing fluid after bleomycin injection.

Methods: Bleomycin (5 mg/kg) was injected into the respiratory tract of Male S-D (Sam:TacN) white rats weighing 250g each for the induction of pulmonary fibrosis. The rats were divided into a bleomycin injection (experimental) group and a non-injection (control) group, which were both measured for weight. The experimental group (9 rats) was injected with 0.3 ml (5 mg/kg) of bleomycin in the respiratory tract, and the control group (3 rats) was injected with the same amount of physiological salt solution, and the remaining 3 rats were not given any injection. After 5 weeks, surfactant protein A in the rat¡¯s bronchial washing was measured using immunoabsorbent assay (R&D System). Finally, the histopathologic changes were confirmed with light microscopic study.

Results: The average weight of the rats was 288.3¡¾12.4 g for the experimental group and 279.8¡¾1.9 g for the control group. But there were no statistical significance (p>0.05). In the study group, the surfactant protein A value had increased in BAL fluid. But there were no statistical significance (p>0.05) because of the sample size. Morphologically, in the light microscopic examination, thickening of interstitium with chronic inflammatory cell infiltration and collapsed alveolar spaces were noted.

Conclusion : In the white rats with pulmonary fibrosis induced by bleomycin injection, surfactant protein A level in the bronchial washing fluid increased. But the increase was beyond the statistical significance because of the sample size. More extensive researches on changes of surfactant protein A in pulmonary fibrosis are needed.
